Neutralization utilized 5 µl of SARS-CoV-2 RVPs in a 384-well plate. GFP positive cells were detected with an Intellicyt iQue flow cytometer using the BL-1 channel (Ex. 488 nm, Em. 530).
Neutralization experiments were conducted using 5µL of SARS-CoV-2 RVPs (Omicron BA.1, INTG Cat # RVP 768G; Omicron BA.2, INTG Cat # RVP 770G; Omicron BA.2.75, INTG Cat # RVP 776G; Delta, INTG Cat # RVP 763G; D614G, INTG Cat # RVP 702G) in a 384-well plate using HEK-293T monoclonal cells stably overexpressing ACE2 (INTG Cat # CHA102).
Infectivity is determined in HEK-293T cells. Data are based on three independent tubes, each tested in quadruplicate.
Flow cytometry analysis of 293T-hsACE2 cells (Cat# C-HA102) with an ACE2 specific monoclonal antibody (R&D Systems Cat# MAB9332-100). Detection using a secondary antibody labelled with AlexaFluor 488 (Jackson ImmunoResearch Cat# 115-545-003). Fluorescence was detected with an Intellicyt iQue flow cytometer using the BL-1 channel (Ex. 488 nm, Em. 530).

Reporter Virus Particles (RVPs) are replication-incompetent pseudotyped virus particles that enable safe (BSL-2), easy, and high-throughput viral infectivity and neutralization assays using standard detection instrumentation. RVPs display antigenically equivalent membrane protein(s) on a heterologous virus core and carry a modified genome that expresses a convenient optical reporter gene within 24 hours of cellular infection.  

SARS-CoV-2 RVPs display antigenically correct spike protein on a heterologous virus core ideal for SARS-CoV-2 neutralization assays.  

C-HA102 is a monoclonal cell line of human embryonic kidney cells (HEK-293T) that overexpress human angiotensin-converting enzyme 2 (hACE2, GenBank Accession# NM_021804.3) to facilitate the binding of SARS-CoV-2 Reporter Virus Particles. This clone stably expresses hACE2 for up to 30 passages under puromycin selection and was selected for its transduction efficiency by SARS-CoV-2 RVPs. The cells are adherent and have an epithelial morphology.  

IM-CoV2-G06 is a recombinant monoclonal antibody that neutralizes early SARS-CoV-2 variants including Wuhan-Hu-1, D614G, Alpha, and Delta. This clone weakly neutralizes Omicron variants BA.1 and BA.1.1. 

VSV RVPs function as a control for RVP experiments to differentiate specific versus non-specific effects on neutralization. VSV RVPs display VSV-G on a heterologous MLV core.
