Neutralizations for DENV1-4(using a monoclonal antibody) utilized 5 µl of DENV RVPs in a 96-well plate. Renilla luciferase activity measured using the Promega Renilla luciferase assay system (Promega #E2810). Sample luminescence was read using a Perkin-Elmer Envision plate reader.
Infectivity for DENV1-4 determined in BHK DC-SIGN cells (Cat# C-BD101).
Flow cytometry analysis of BHK DC-SIGN cells (Cat# C-BD101) with a DC-SIGN specific monoclonal antibody (R&D Systems Cat# MAB161). Detection using a secondary antibody labelled with AlexaFluor 488 (Jackson ImmunoResearch Cat# 115-545-003). Fluorescence was detected with an Intellicyt iQue flow cytometer using the BL-1 channel (Ex. 488 nm, Em. 530).

Reporter Virus Particles (RVPs) for all four dengue serotypes are replication-incompetent virus-like-particles that enable safe (BSL-2), easy, and high-throughput viral infectivity and neutralization assays using standard detection instrumentation.

DENV RVPs are used to test the ability of serum, antibodies, and drugs to neutralize infectivity. DENV RVPs are structurally intact dengue virus-like particles, capable of a single round of infection and carry a genome that expresses a luciferase optical reporter gene upon infection. RVPs are derived from a baby hamster kidney cell line (BHK-21) and are produced using DENV CprME structural genes and a Flavivirus reporter replicon. 

C-BD101 is a monoclonal cell line generated using BHK baby hamster kidney cells to create a permissive cell line for infection by flavivirus Reporter Virus Particles. The cells were developed from BHK21 clone 15 (Center for Vector-borne Diseases, UC Davis). The cells are adherent and have a fibroblast morphology.  

Cells are intended for research purposes only. Protocols are provided for neutralization and infectivity assays.
