HA assay was performed using 1% Guinea Pig RBCs in a 96-well plate. The HA assay was used to determine an Influenza A TiterSafe titer at 4 HA units. The titer at 4 HA units and hyperimmune serum were used to perform the HAI assay. Reported serum dilutions are defined prior to TiterSafe volume addition.
HA and HAI assays were performed using 1% Guinea Pig RBCs in a 96-well plate. Neuraminidase maintains enzymatic activity that may interfere with hemagglutination at concentrations used in HA, but not HAI assays (HA assay, column 1).

Influenza TiterSafe, an HAI reagent, consists of non-replicative virus-like particles (VLPs) that display influenza HA and NA on a lentivirus core. TiterSafe replaces live virus in the hemagglutination inhibition assay (HAI), the standard assay for testing immune sera to determine to assess serum antibody titers. Protocols are provided for HA and HAI assays, with guinea pig RBCs the recommended target.
